Le prove d’esame consistono in:

prova scritta con quesiti a risposta aperta, che potrà vertere sui seguenti argomenti:

  • spettrometria di Massa in Alta e Bassa Risoluzione, compresi diversi tipi di combinazioni degli analizzatori di massa maggiormente usati per applicazioni Omiche sia in ambito quantitativo che qualitativo anche con riferimento alla modalità di acquisizione (DDA, DIA e altro) compresa la mobilità ionica e il suo impiego;
  • la cromatografia sia in fase GAS che in fase liquida (HPLC e UPLC) compresi i sistemi di arricchimento on-line e in SPE e le modalità di preparazione delle matrici biologiche in campo clinico con particolare riferimento all’approccio metabolomico e lipidomico;

  • disegno sperimentale, registrazione dei dati relativi ai campioni e la loro conservazione. Preparazione delle matrici biologiche e costruzione delle sequenze analitiche in Metabolomica e Lipidomica. Valutazione e verifica della riproducibilità analitica (RT) e dell’errore di misura del m/z e del CCS. Utilizzo di diverse modalità di acquisizione sia in MS, MRM, DDA che in DIA con l’uso della Mobilità Ionica;
  • gestione dei controlli di qualità interni ed esterni per la determinazione quantitativa dei vari pannelli metabolici (per esempio: aminoacidi, acilcarnitine, acidi Biliari);
  • utilizzo di spettrometri di massa (Waters) a bassa ed alta risoluzione con mobilità ionica, interfacciati con HPLC, UPLC e GC;
  • uso del Software MassLynx per la gestione strumentale e i suoi applicativi con particolare riferimento a: TargetLynx, MSeDataViewer, DriftScope, NeoLynx e FractionLynx;
  • lettura e interpretazione dati in Microsoft Excel, SIMCA, e Progenesis QI.

Verrà accertata la conoscenza della lingua inglese (livello di riferimento B2), tramite lettura e traduzione testo tecnico scientifico.

Project Humanize: A Step Towards Creating a Heart from Pluripotent Stem Cells

Organ transplantation represents the ultimate solution for numerous serious diseases, but the availability of organs is sadly insufficient to meet global demands. In Italy alone, in 2024, there are approximately 8,000 patients on the waiting list, a situation that makes the search for alternative sources of organs urgent. One of the most promising solutions to address this issue could lie in the production of human organs from pluripotent stem cells. These cells have the ability to differentiate into any type of tissue or organ and, thanks to their capacity to multiply in the laboratory, could allow for the large-scale generation of human organs and tissues.

The Humanize project, funded by the Spanish foundation La Caixa with 1 million euros for three years, aims to develop technologies capable of identifying and producing human stem cells with high potential to generate organs. The research is focused on the human heart, as the shortage of donors for this organ is one of the most critical issues in the medical field. The principal investigators of the project, which focuses on the generation of a human heart, are Graziano Martello, professor in the Department of Biology at the University of Padua and director of the Pluripotent Stem Cell Biology Laboratory (Lab Martello), and Xabier Lopez Aranguren, researcher at Cima Universidad de Navarra in Spain.

Previous studies based on rodents have shown that pluripotent stem cells can generate entire organs, such as the heart and pancreas, in animals that, due to genetic defects, would not be able to generate them. These studies have also highlighted that not all rodent stem cells are capable of forming organs.
The goal of the Humanize project is to identify human stem cells with similar potential, capable of developing functional organs. The next step will be to verify, through animal models, whether the produced human stem cells can effectively contribute to the formation of organs. The verification of the project's underlying hypothesis, which will be experimentally tested in Spain, will focus on animals such as pigs, whose organs are already used for heart valve transplantation.

The impact of this research would not only improve the quality of life for patients awaiting transplants but also benefit the global healthcare system. If confirmed, the potential to produce human organs from pluripotent stem cells could solve the problem of donor scarcity and reduce the costs associated with treating patients on transplant waiting lists. Additionally, the development of technologies for human organ production would open new opportunities in the biotechnology sector, stimulating the creation of new companies and productive activities in a high-demand sector.

After the success in Padua of the first transplant of a controlled non-beating heart last year, the news of December 11, 2024, is of the world's first heart transplant from a non-beating donor to a fully beating heart, again in Padua. The feat was accomplished by the team from the UOC Cardiac Surgery Department of the University Hospital of Padua, led by Gino Gerosa, professor at the University of Padua.

This revolution will improve the outcomes of heart transplants from non-beating donors by avoiding controlled heart arrest, eliminating ischemia and reperfusion damage both during extraction and transplantation, ensuring a quicker recovery of heart function, and enhancing post-operative cardiac performance.

"Gino Gerosa and his extraordinary team have, for the first time in the world, crossed a new frontier in cardiac surgery and transplantation. A new indelible page in the international history of this discipline has been written in Veneto. There are no words left to express the esteem and gratitude to Professor Gerosa, all the members of his team, and the University Hospital of Padua," said the President of the Veneto Region, Luca Zaia, in celebrating the world's first beating heart transplant, which took place in Padua.

The date of November 14, 1985, in Padua, when the cardiac surgery team led by Vincenzo Gallucci successfully completed Italy's first heart transplant, is now a part of history.
